姜黄素减轻小鼠癫痫发作

摘要: 目的 探讨姜黄素对小鼠癫痫发作的作用及机制。 方法 C57 / BL6 雄性小鼠 60 只,随机分为 3 组:正常组、 癫痫模型组和姜黄素治疗组,每组 20 只。 观察记录每组小鼠癫痫发作 Racine 分级评分和癫痫发作的潜伏期。 免 疫荧光染色检测海马 CA3 区 GFAP 和 IBA-1 表达,蛋白免疫印迹(Western blot)检测海马 IL-1β、NLRP3、caspase1 的 表达水平。 结果 姜黄素治疗组小鼠癫痫发作 Racine 分级评分显著低于癫痫模型组( P< 0. 05) ,发作潜伏期显著 延长(P<0. 05) 。 与正常组相比,模型组海马 CA3 区 GFAP 和 IBA-1 的表达显著增加(P<0. 01) ,胶质细胞呈现明显 激活状态,IL-1β、NLRP3、caspase1 蛋白的相对表达量显著升高( P< 0. 01) ,与模型组相比,姜黄素组海马 CA3 区的 GFAP 和 IBA-1 阳性细胞数显著减少 ( P < 0. 05) ,海马 IL-1β、NLRP3、 caspase1 蛋白相对表达量也显著降 低 ( P < 0. 05) 。 结论 姜黄素可以减轻癫痫发作程度,这可能通过调节 NLRP3 / caspase1 通路抑制海马炎性反应来实现。

Abstract: Objective To explore the effect and mechanism of curcumin on seizures in mice. Method 60 C57 / BL6 male mice were randomly divided into normal group, epilepsy model group and curcumin treatment group, with 20 mice in each group. Racine grade score of seizures and the latency of seizures in each group were observed and recorded. The expressions of GFAP and IBA-1 in hippocampal CA3 region were detected by immunofluorescence staining, and the expression levels of hippocampal IL-1β, NLRP3, caspase1 were detected by Western blot. Result Racine score of seizures in curcumin treatment group was significantly lower than that in epilepsy model group(P< 0. 05) , and the seizure latency was significantly prolonged ( P < 0. 05 ) . Compared with the control group, the expression of GFAP and IBA-1 in hippocampal CA3 region in the model group was significantly increased ( P < 0. 01) , glial cells were significantly activated, and the expression levels of hippocampal IL-1β, NLRP3, caspase1 were upregulated significantly ( P < 0. 01 ) . Compared with the model group, the number of GFAP and IBA-1 positive cells in hippocampal CA3 of curcumin group were decreased dramatically( P< 0. 05) , and the expression levels of hippocampal IL-1β, NLRP3, caspase1 were decreased notably ( P < 0. 05) . Conclusion Curcumin can reduce the severity of seizures, which may be through the regulation of NLRP3 / caspase1 to inhibit hippocampal inflammation.
